Another method of creating a genomic library ia to use bacteriophage. Bacteriophages can package pieces of genomic DNA from a cell and then infect other cells. Each plaque on a plate will be a separate piece of genomic DNA.

Recombinant phage DNA library

cDNA or complementary DNA can be used to make a genomic library. Instead of using genomic DNA, researchers can use messenger RNA (mRNA) to create a library. The cDNA is the complement of that RNA. Because cDNA is reverse transcribed from mRNA, cDNA libraries contain only expressed genes in the genome which can be important information for researchers. These cDNA can be replicated using DNA Polymerase.

Restriction-enzyme digests can be used to chop up a cell's genomic DNA into fragments and ligate the plasmids. These can be transformed into bacterial cells which can be grown out. Each cell will have different clones which have different fragments of the genomic DNA.
