Conjugation can be used in molecular cloning experiments when a cell with the F plasmid and the recombinant DNA can mate with cells that do not have the F plasmid. Through this process, the plasmids can be transferred directly to other cells and the cell can act as a donor to other cells.

During conjugation in E.coli, the conjugation pilus of the F+ cell comes into contact with the F- cell that does not contain the F plasmid. A single stranded copy of the F plasmid is transferred between the two cell. The F plasmid is then synthesized in the recipient cell to make it a F+ cell.
