Endospore staining is a differential technique that uses two stains to distinguish endospores from the rest of a bacterial cell. The most commonly used method, the Schaeffer-Fulton technique, uses heat to push a primary stain (malachite green) into the endospore. After washing with water—which decolorizes the rest of the cell while the endospore retains the green stain—a secondary counterstain (safranin) is applied to color the vegetative cells pink. When viewed under a microscope, green endospores appear either within or separate from the pink vegetative cells, while specimens without endospores will only show pink cells.

Sporulation is the process by which vegetative bacterial cells transform into dormant endospores, typically initiating when nutrients become depleted or environmental conditions become unfavorable. The process involves asymmetrical cell division to form a forespore, followed by the sequential addition of protective layers, culminating in the release of the mature endospore upon the disintegration of the mother cell.

While not all bacteria possess the ability to form endospores, several clinically significant, gram-positive pathogens belong to the endospore-forming genera *Bacillus* and *Clostridium*. These include *Bacillus anthracis* (anthrax), *Clostridium tetani* (tetanus), *Clostridium perfringens* (gas gangrene), and *Clostridium botulinum* (botulism), as well as the related pathogen *Clostridioides difficile* (pseudomembranous colitis). These pathogens are particularly challenging to combat medically because their endospores are highly resilient and extremely difficult to eradicate.
