Identification of immunogenicity of Self-amplifying RNA SARS-CoV-2 lipid nanoparticle vaccine candidate induces high neutralizing antibody titers in mice
- IFN-γ ELISpot kits were used to assess IFN-γ T cell response.
- Semi-quantitative ELISA was used to observe antigen-specific IgG, IgG1 and IgG2a titers in mouse serum.
- Neutralization ability of vaccinated animals’ and recovered patients’ serums against SARS-CoV, SARS-CoV-2, MERS-CoV, and 229E-CoV were assessed using HIV-pseudotyped luciferase-reporter based system.
- The ability for the serum of vaccinated animals to neutralize wild type SARS-CoV-2 virus was observed using a neutralization assay on Vero-E6 cells.
- Cytokine response fo splenocytes and serums of each mice were quantified using a custom 25-plex ProcartaPlex Immunoassay (ThermoFisher Scientific, UK) on a Bio-Plex 200 System (Bio-Rad).
