N proteins are recruited to RTCs via binding to nonstructural protein 3 (nsp3), a component of the RTC. N proteins mainly, but not exclusively, bind to nsp3 among the other nonstructural proteins. Like every protein, nsp3 has an N- and C- terminus. N proteins interact specifically with the N-terminal region of nsp3.

In infected cells, N proteins must be recruited to replication-transcription complexes (RTCs) for RNA synthesis to proceed. RTCs are sites of RNA synthesis and are complexes made up of nonstructural proteins (nsp).
