454 Sequencing is a type of next generation sequencing where a DNA sample is made into single strand fragments. The fragments are adapted to  bind to a bead. Each bead allows for isolating the DNA to be amplified by PCR and then sequenced. During sequencing probed nucleotides when annealed to DNA send a light to a detector to sequence the DNA. This allows for rapid DNA sequencing.

OpenStax Microbiology Textbook

The Sanger DNA sequencing method was developed by Frederick Sanger in 1972. It uses dideoxynucleotides that are when incorporated into DNA, terminate the amplifying strains. This creates strands of DNA that when run on an agarose gel, show a DNA ladder that maps the original DNA sequence. These fragments can also be read on a fluorescence detector.
