Gram-positive and gram-negative bacteria exhibit distinct flagellar basal body configurations due to structural cell wall variations. In both types, a hook connects the external filament to a rod that links to the basal body in the inner membrane, which consists of a C-ring, an MS-ring, stators, and a type III secretion protein. Because gram-negative bacteria have an outer membrane and a periplasmic space, their flagellar configuration additionally includes an L-ring in the outer membrane and a P-ring in the cell wall.

Arrangement of Flagella

Flagella staining is a specialized technique used to visualize flagella under a light microscope. Because flagella are extremely thin, the procedure first applies a mordant—typically tannic acid or potassium alum—to coat and thicken the flagellum. Following the mordant, the specimen is stained with a dye, most commonly pararosaniline or basic fuchsin, rendering the thickened flagella visible.
