TEM Specimen Dehydration and Embedding

Because biological cells are too soft to be cut into the extremely thin sections required for a transmission electron microscope (TEM), they must be embedded in plastic resin. This involves dehydrating the cells through a series of ethanol soaks so the ethanol replaces the cellular water. The resin then dissolves in the ethanol, enters the cell, and solidifies to provide the structural support needed for cutting without damage.
