Designing an Experiment to Test for Altruism

Imagine you are an economist studying behavior in a one-shot, two-player game where each player can either 'Cooperate' or 'Defect'. The standard setup shows that a significant minority of players choose to 'Cooperate', even though 'Defect' is the individually rational choice. A leading hypothesis is that this cooperation stems from altruism. Propose one specific modification to the experimental design that would help you test whether altruism is the true motivator, as opposed to other potential factors like player confusion or a belief that they might interact with their opponent again.
