Gram Stain

In multi-step staining techniques, a primary stain is the initial dye applied to a specimen. It serves to impart a base color to all targeted cells or structures before any subsequent differentiating chemicals or secondary stains are used.

Primary Stain

A mordant is a chemical substance used in staining procedures to set or stabilize stains or dyes. It typically functions as a trapping agent that forms a complex with a primary dye, causing it to clump and remain securely contained within specific cellular structures.

A decolorizing agent is a chemical solution used in differential staining methods to selectively wash away the primary dye from certain cells or structures. Its selective action allows for the visual differentiation of organisms based on their structural abilities to retain or lose the initial stain.

Decolorizing Agent

A secondary counterstain is an additional dye applied after a decolorization step in a differential staining procedure. It is used to impart a contrasting color specifically to the cells that have been decolorized, making them visible and easily distinguishable from the cells that retained the primary stain.

Secondary Counterstain

Acid-fast staining is a commonly used differential technique that distinguishes between two types of gram-positive cells: those possessing waxy mycolic acids in their cell walls and those lacking them. In this procedure, carbolfuchsin is applied as the primary stain. Waxy, acid-fast cells retain this red or pink dye even after an acid-alcohol decolorizing agent is applied, whereas non-acid-fast cells lose the primary stain and are subsequently rendered blue by a methylene blue secondary counterstain.

Acid-Fast Stain

Because a protective outer capsule does not absorb most basic dyes, a negative staining technique is typically used for capsule staining to determine if cells in a specimen are encapsulated, which serves as an important diagnostic tool for evaluating a microbe's virulence. In a negative capsule stain, a dye such as India ink or nigrosin colors the background without penetrating the capsule, leaving a visible clear halo around each cell without requiring the specimen to be heat-fixed. Alternatively, a combination of positive and negative staining techniques can be used, where a positive stain colors the body of the cell and the negative stain colors the background, rendering the capsule as a clear halo.

Capsule Stain

Endospore staining is a differential technique that uses two stains to distinguish endospores from the rest of a bacterial cell. The most commonly used method, the Schaeffer-Fulton technique, uses heat to push a primary stain (malachite green) into the endospore. After washing with water—which decolorizes the rest of the cell while the endospore retains the green stain—a secondary counterstain (safranin) is applied to color the vegetative cells pink. When viewed under a microscope, green endospores appear either within or separate from the pink vegetative cells, while specimens without endospores will only show pink cells.

Flagella staining is a specialized technique used to visualize flagella under a light microscope. Because flagella are extremely thin, the procedure first applies a mordant—typically tannic acid or potassium alum—to coat and thicken the flagellum. Following the mordant, the specimen is stained with a dye, most commonly pararosaniline or basic fuchsin, rendering the thickened flagella visible.

Differential staining is a technique that uses multiple different stains to distinguish between organisms based on their varying interactions with the dyes. Unlike simple staining, this method can cause different organisms within the same sample to appear in different colors, making it highly useful in clinical settings for identifying specific pathogens.

This technique generally causes all of the organisms in a sample to appear uniformly as the same color, even if the sample contains multiple types of organisms, making it useful for basic visualization but not for differentiation.
